Wet Lab

CRYOSTAT LABORATORY
Sample preparation, fixation, cryoprotection, freezing of the heads and brains, and other wet bench activities are carried out in the LONI Wet Lab. Equipment present in the lab includes a peristaltic pump for perfusion, equipment for embedding samples, common laboratory instruments (PH meter, balances and chemistry equipment), and a chemical hood. A modified cold room contains a Leica CM3050S Cryostat, image capture equipment and a PC controlled camera system networked to the file servers. Modifications to the cryostat include a camera mount built to accept a Polaroid DMCIe high-resolution digital camera with an Optem macro lens and a Hiedenheim micrometer that allows the blockface to be returned to its original position (within 0.5um).
 Additionally, the lab also contains an American Optical model 860 sliding microtome for non-frozen sectioning. The microtome has also been modified with a camera mount to accept a Polaroid DMCIe high-resolution digital camera.

 A complete histology laboratory is available also with full complement of staining accessories and an Olympus AX70 research-grade light microscope. The microscope is capable of both high-magnification and low-magnification (macro) imaging and is attached to a PC controlled camera system (Nikon DXM-1200 high-resolution digital camera) networked to the file servers.

CRYOMACHROTOME LABORATORY:
 Fixing and cryoprotecting the heads and brains and other wet bench activities are carried out in the Wet Lab. Equipment present in the lab include common laboratory instruments (PH meter balances and chemistry equipment), and a hood. A modified cold room contains the PMV Cryomacrotome and histological setup. The machine has a PC controlled camera system networked to the file servers.
